What is metabolomics?

Metabolomics refers to the field of study that comprehensively analyzes the various metabolites synthesized and consumed within living organisms, as well as its analytical techniques. By investigating how metabolites such as low-molecular organic acids and amino acids are metabolized in living organisms, it is possible to detect specific diseases early in biological samples such as plasma and serum, investigate the efficacy of medicines, and improve food products. It is applied to sample quality control, etc.

Solid-Phase Derivatization

In metabolomics, it is desired to comprehensively measure many substances at once in order to understand the metabolism of all substances. Metabolites such as amino acids, organic acids, and sugars are highly polar and nonvolatile substances, so they are usually the target substances for LC measurement. However, since they each have different physical properties, it is difficult to analyze them all at once using LC. Therefore, by derivatizing these non-volatile substances with different physical properties, we change them into volatile substances and perform simultaneous analysis using GC. In this way, simultaneous GCMS analysis using "derivatization" of amino acids, organic acids, and sugars is important because it allows comprehensive analysis.

Solid-Phase Derivatization

Conventional pretreatment for "derivatization" takes time for the freeze-drying (dehydration) and derivatization reaction steps, and there are many manual steps such as extraction, which are complicated and time-consuming, and require analytical skills. The current situation is that the data may vary depending on the situation.
Compared to conventional derivatization reactions, the solid-phase derivatization method allows reactions to be carried out in a shorter time, with higher efficiency, and with less solvent, so it has advantages such as improved reproducibility and reduced costs.


In addition, by combining it with the online SPE-GC system "SPL-M100", it is possible to automate everything from preprocessing to measurement.
Furthermore, since the entire amount of eluate from the solid phase can be introduced into GC/MS, higher sensitivity can be expected.
By changing the derivatization reagent, it is possible to efficiently analyze different targets.

Automatic analysis by online SPE-GC system

One of the advantages of using solid phase extraction for pretreatment is that it is easy to automate the pretreatment operation.
Furthermore, by using an online SPE-GC system that can inject the entire amount of eluate from the solid phase, it is possible to perform fully automated analysis from pretreatment after extraction/protein removal to measurement.
It minimizes the differences in individual skills and laboratories that have been an issue with conventional analytical methods, and improves the precision and reproducibility of analysis.
